Accurate
prediction of safety remains a major challenge in drug
development. Common
safety problems that arise include cardiotoxicity,
hepatotoxicity and nephrotoxicity. In
vitro
tests are routinely run in preclinical development in an attempt
to rapidly eliminate unsafe compounds and promote promising
potential candidates. Progress
has been made in developing higher-throughput techniques for the
early detection of compounds causing problems in the heart,
liver and kidney. When it comes to in
vivo screening one of the major challenges is accurately
predicting human drug toxicity with nonhuman animal models.
Preclinical
Drug Safety 2009
will evaluate the in
vitro &
in vivo toxicity screens in use today, assess how far
computational tools have come in helping to predict compound
toxicity and review imaging technologies used in safety
prediction. Some discussion will be included on idiosyncratic
and mitochondrial toxicity. A full session will be dedicated to
efforts being made to improve predictive
preclinical models
with special mention of species (e.g. rat-to-man) issues.
